I've read the original book. Is that a problem?
You mean, is the culprit the same person as it was in the book, the play, or the movie? Not necessarily. That's the beauty of a mystery with this many characters; the villain can be anybody. So play the game content in the knowledge that you get to figure it all out again.
On the other hand, you might find it helpful to know the general story, since there are a number of plot points and so on that aren't explained in detail in the game. And since the book is so handily included with the game (at least in the early versions), it's right there for you to refer to.
