One is to find and scribble down a copy of the fifth and final evil red symbol.
- Okay. (Sigh!) Where's the evil red symbol for this one?

The other is to get whatever Dr. Hermann has clutched so tightly in his right hand.
- Dr. Hermann has something clutched in his right hand. How do I get it?

Once you're through with that, Detective Collier will show up and chase Samuel off. Samuel will then return to the sanatorium with the notion of getting some of James' hairs for comparison to the ones taken from Dr. Hermann's hand.
