Exploring the levels
Obviously, the best tactic is knowing the level layout. Play the level beforehand. Read the individual level hints in this file. You can find locations of weapons, which will definitely help you in defeating your opponent.
Another thing to note is that any items you pick up will leave behind a Duke Nukem 3D symbol (radioactive sign) that corresponds your color. This can give your general location away, since your opponent would know that you were in the area within the last 30 seconds.
Additionally, if you have a good sound card, you should be able to tell whether someone is close by. The louder of the sounds, the closer he is. For this reason, I don't recommend making Duke "speak" (like he would do in front of a mirror). This can give your location away, something you probably won't want to do. Or maybe you do, case depending.
There are many cracked walls and doors you find in some levels that "connect" various parts. For instance, in Hotel Hell (E3L8) there's a crack right at the end of the level that will lead right to the beginning. Although this is useless in single player, it will help getting from place to place. Occasionally you'll also find transporters around, so getting from beginning to end of the level can be fast and easy.
