Main Quests
Fallout: New Vegas is a very large, nonlinear game in pretty much the same way that Fallout 3 was. There is a main plotline, which is defined by the series of quests listed below. You start off small (as a left-for-dead courier) and end up shifting the entire region's political balance of power. There's no time limit on getting or doing any of the quests, so once you finish the character creation process, feel free to explore each region you come to, do misc. and faction quests, etc. You pretty much have to in order to build up your character to where he can finish the main quests, anyway.
