Do I have to play the other seasons first?
You don't need to for the sake of the plot, no; like the first two Sam & Max seasons, this one begins an entirely new story arc. You'll get a lot more out of it (and catch a lot more of the jokes) if you have played the first two seasons, of course -- but if you've been on Mount Everest for the last three years, feel free to start with this one.
